Portable single-cell analysis with the micropeek smartphone adapter and COC microfluidic chips

We report a single-cell analysis platform using smartphone-controlled fluorescence microscopy to enumerate and monitor particles and immobilized cells in cyclic olefin copolymer (COC) microfluidic chips. The crowdfunded μPeek smartphone adapter is equipped with multiple LEDs, an integrated motorized lens, and is capable of bright field, dark field and fluorescence microscopy down to a resolution of ~ 1 μm. We combined this compact, portable imaging platform with a custom Android App and low-cost microfluidic chips for single-cell analysis. The COC microfluidic chips are produced via lithography, electroplating, and thermal imprinting (LIGA). We successfully applied the device for particle counting, fluorescent single cell enumeration of white blood cells in PBS and whole blood, and monitored the response of single MCF-7 cells to oxidative stress.
