Living in a bubble: On chip monitoring of microbial production in lipid vesicles
(2019)23rd International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2019)We present a novel microbial cultivation strategy combined with a microfluidic platform for real-time monitoring of microbial growth and production. Escherichia coli overexpressing the riboflavin (vitamin B2) biosynthetic pathway is encapsulated in giant lipid vesicles (GUVs) and cultivated on-chip. The behavior of cultures under optimum conditions and under antibiotic stress are compared.Other Conference Item -
Identifying extracellular vesicle populations from long-term cultured single cells using multi-color TIRFM
(2019)23rd International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2019)We here report a double-layer microfluidic device for detecting and identifying secreted extracellular vesicle populations, among them exosomes, from single long-term cultured cells using multi-color Total Internal Reflection Fluorescence Microscopy (TIRFM). Using a custom-developed MATLAB script for image processing we achieved quantitative population analysis of vesicles secreted from single Michigan Cancer Foundation (MCF)-7 cells ...Other Conference Item -

Tubing-free Microfluidic Platform for Co-culture of 2D Adherent Cells and 3D Microtissue Spheroids
(2019)23rd International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2019)We present a tubing-free, standard 384-well-format-based microfluidic platform to co-culture 2D adherent cells and 3D microtissue spheroids, while they are fluidically interconnected. The culture wells of the platform are separated by capillary stop valves and can be selectively treated to promote or to prevent adhesion of cells and/or microtissue spheroids. The wells can be interconnected after surface treatment by simply filling the ...Other Conference Item -
Microfluidic system for cultivation and monitoring of individual riboflavin overproducing Escherichia coli cells
(2019)23rd International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2019)We present a microfluidic platform allowing real-time observation of heterogeneous bacterial populations for basic studies of microbial growth and production. We succeeded in batch culturing and monitoring Escherichia coli cells overexpressing the riboflavin (vitamin B2) biosynthetic gene cluster which enables them to secret preparative amounts of riboflavin. Production in cultures with and without induction of the gene cluster were compared.Other Conference Item -
Absorbance spectra-activated droplet sorting for high-throughput label-free chemical identification
(2019)23rd International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2019)We introduce the Absorbance Spectra-Activated Droplet Sorter for high-throughput label-free chemical identification of droplets and subsequent sorting. The 200 nm to 1050 nm droplet absorption spectra - with 0.4 nm resolution - is detected and analyzed up to 2100 spectra per second with a custom software. Our system combines an optimized design and UV-VIS absorbance down to 200 nm to quantitatively measure unlabeled proteins at 10 μM and ...Other Conference Item -
Passive wireless sensing of microtissue properties
(2019)23rd International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2019)Passive wireless sensing devices [1] make use of an interrogation technique that does not require powersupply or wire-connections to the sensor device, which provides large configuration freedom. A prototype passive wireless sensor, together with the corresponding readout platform, is proposed here. The sensor is a square split ring that responds to impedance changes of microtissue spheroids. The wireless sensing system is used to detect ...Other Conference Item -
A multiplexed cell-free assay in double emulsion droplets
(2019)23rd International Conference on Miniaturized Systems for Chemistry and Life Sciences (MicroTAS 2019)We introduce a novel on-chip assay, performed in water-in-oil-in-water double emulsions, that uses cell-free protein synthesis and artificial liposomes for the screening of membrane disrupting toxin DNA libraries according to their antimicrobial activity and host membrane safety. Our method represents a unique approach to the selection and potentially directed evolution of membrane-disrupting toxins.Other Conference Item -

Double machine learning methods: Beyond independence
(2022)Oberwolfach Report ~ Re-thinking High-dimensional Mathematical Statistics (Workshop Report 2022, 25)Double machine learning is a tool to combine machine learning algorithms and statistical models to estimate and make inference on low-dimensional parameters. These parameter estimators converge at the parametric rate and follow a Gaussian distribution asymptotically. We present three use cases: estimating the linear part in a partially linear endogenous model and in a partially linear mixed-effects model and treatment effect estimation ...Other Conference Item
