MALDI Mass spectrometry on high-density droplet arrays: Matrix deposition, selective removal and recrystallization

Abstract
Mass spectrometry is a powerful, label-free method to analyze high-density droplet arrays. Here, we introduce a new workflow for sample preparation of thousands of droplets, which significantly improves the detection by matrix-assisted laser desorption/ionization-mass spectrometry (MALDI-MS). We deposited matrix compounds in a custom-made sublimation chamber followed by a recrystallization step and achieved increased signal intensities for a model protein. Furthermore, selectively removing the matrix before recrystallization enhances spatial resolutions alongside the ionization efficiency. Mehr anzeigen
