A Pipeline for Personalized Brain Modeling: From Stimulation to Network Activity

Abstract
Neurological and psychiatric disorders are the leading cause of disability worldwide. Personalized computational modeling is a powerful method for probing whole brain network dynamics in both health and disease, and for creating optimized brain stimulation-based therapies. To this end, we developed an automated patient-specific modeling pipeline starting from structural magnetic resonance imaging (MRI) scans, and resulting in a 16 tissue, biophysically realistic 3D head model functionalized with a large-scale dynamic model of brain activity. We simulated this model subject to various electrical brain stimulation (EBS) modalities using Sim4Life’s (ZMT Zurich MedTech AG, Switzerland) electroquasistatic solver to determine electromagnetic (EM) field distributions in each tissue. The impacts of EM exposure on network dynamics were evaluated using The Virtual Brain (TVB) software for personalized brain network and signal modeling. Finally, TVB brain network activity simulations were translated algorithmically into predictions of macroscale brain signals. The preceding steps were integrated into an automated pipeline on o2S2PARC, an open, cloud-based platform for creating, sharing, and publishing computational models and data analysis workflows.
